Panasonic VCR without remote

We recently picked up a second-hand PV-V4521. Is there any way to
bring up on-screen display without a remote (the only PANA-compatible
RC we have is a Program Director VSQS1250, which AFAIK is not
compatible)

You need an LSSQ0264 remote. I see them for AU$3 (buy it now price) on
eBay.

You're going to need a compatible remote.

Most Panasonic VCRs use similar remote controls, we have had quite a few
over the years and freely interchange the remotes among them with no problem.

The exception is a PV-4662 VCR I picked up for a few bucks at a thrift
store -- this model requires a remote with thumbwheel built in for some of the
menu functions. Unless yours falls into this category you should be able to
use just about any Pansonic VCR remote that you might find at a thrift store,
yard sale, flea market, etc.
